This study was designed to determine the effect of Mangifera indica flowers decoction, on the acute and subacute models of induced ulcer in mice and rats. A single oral administration of the aqueous decoction (AD) from M. indica up to a dose of 5 g/kg, p.o. did not produce any signs or symptom of toxicity in the treated animals. The oral pre-treatment with AD (250, 500 and 1000 mg/kg) in rats with gastric lesions induced by ethanol, decreased the gastric lesions from 89.0 +/- 6.71 (control group) to 9.25 +/- 2.75, 4.50 +/- 3.30 and 0, respectively. Pretreatment with AD (250, 500 and 1000 mg/kg) to mice with HCl/ethanol- or stress-induced gastric lesions resulted in a dose-dependent significant decrease of lesion index. In the piroxicam-induced gastric lesions, the gastroprotective effect of AD was reducing with the increase of the AD dose. In the pylorus-ligature, AD (p.o.) significantly decreased the acid output indicating the antisecretory property involved in the gastroprotective effect of M. indica. Treatment with AD during 14 consecutive days significantly accelerated the healing process in subacute gastric ulcer induced by acetic acid in rats. Pretreatment with N-nitro-L-arginine methyl ester (L-NAME), an inhibitor of NO-synthase, did not abolish the gastroprotective effects (99% with saline versus 80% With L-NAME) of AD against ethanol-induced gastric lesions. Pretreatment with N-ethylmaleimide (NEM), a blocker of endogenous sulphydryl group, significantly abolished the protective effects of AD against ethanol-induced gastric ulcers (95% with saline versus 47% with NEM). Phytochemical screening showed the presence of steroids, triterpenes, phenolic compounds and flavonoids. Estimation of the global polyphenol content in the AD was performed by Folin-Ciocalteu method and showed approximately 53% of total phenolic on this extract. These findings indicate the potential gastroprotective and ulcer-healing properties of aqueous decoction of M. indica flowers and further support its popular use in gastrointestinal disorders in Caribbean. The present study aimed at evaluating the behavior of the soybean oil with the addition of lemon seed extracts (Citrus limon), in different concentrations, through oxidative stability and also at measuring the antioxidant activity using the DPPH free radical method and total phenolic compounds. The concentration of 2,400 mg.kg(-1) for the lemon seed extract, galego variety, was the one that provided the soybean oil with the best oxidative stability. The maximum antioxidant activities and the concentration values of the natural extract sufficient to obtain 50% of maximum effect, estimated at 100% (EC(50)), determined by DPPH for the extract and gallic acid were 70.58%, 69.94 mu g.mL(-1) and 75.07%, 64.73 mu g.mL(-1), respectively. The concentration of total phenolic compounds, determined by the Folin-Ciocalteu method, was of 76 mg.g(-1). Thus, it can be said that the lemon seed extract, galego, presents natural antioxidant action demonstrating potential to be used in foods.

Four different assays (the Folin-Ciocalteu, DPPH, enzymatic method, and inhibitory activity on lipid peroxidation) based on radically different physicochemical principles and normally used to determine the antioxidant activity of food have been confronted and utilized to investigate the antioxidant activity of fruits originated from Brazil, with particular attention to more exotic and less-studied species (jurubeba, Solanum paniculatum; pequi, Caryocar brasiliense; pitaya, Hylocereus undatus; siriguela, Spondias purpurea; umbu, Spondias tuberosa) in order to (i) verify the correlations between results obtained by the different assays, with the final purpose to obtain more reliable results avoiding possible measuring-method linked mistakes and (ii) individuate the more active fruit species. As expected, the different methods give different responses, depending on the specific assay reaction. Anyhow all results indicate high antioxidant properties for siriguela and jurubeba and poor values for pitaya, umbu, and pequi. Considering that no marked difference of ascorbic acid content has been detected among the different fruits, experimental data suggest that antioxidant activities of the investigated Brazilian fruits are poorly correlated with this molecule, principally depending on their total polyphenolic content. © 2013 Elena Gregoris et al.

There are cassava varieties that present compounds as carotenoids, beta-carotene, lycopene and minerals important for human and animal health. The present study evaluated the antioxidant activity of the white, yellow and pinkish varieties of Manihot esculenta, by mean of the DPPH test and by the ferrous ion-chelating activity. Furthermore, the total phenols, carotenoids, beta-carotene, lycopene and zinc contents were also determined. Utilizing the DPPH test it was possible to find that extracts of boiled samples presented higher antioxidant activity (89.53% -pinkish) in comparison to the fresh samples (1.97% -white). For the ferrous ion-chelating test, the highest activity was found for the boiled pinkish variety extract (63.43%) and the lowest was for fresh yellow extract (17.34%) the white sample did not present activity. The highest concentration of total phenols and zinc content was obtained for the boiled pinkish variety extract 136.12 mg EAG/g of extract and 0,811ppm, respectively, in the concentration of 1000 mu g/mL. The pinkish variety presented also higher quantity of pigments, including carotenoid (29.40 mu g/g), beta-carotene (9.14 mu g/100g) and lycopene (68.92%). According to the results obtained in this study it was possible to conclude that the yellow and pinkish varieties of M. esculenta present quantity of phenolic compounds and minerals sufficient to attribute the antioxidant activity and may thus contribute to reduce oxidative damage and be used as nutraceuticals or directly ingested in the diet to maintain good health.

Ultimamente várias áreas da ciência vêm se unindo a fim de compreender, elucidar, retardar e reverter o processo do envelhecimento da pele. Tal processo deve ser estudado, pois as mudanças no aspecto exterior podem revelar disfunções que ocorrem internamente, além de trazer conseqüências psíquicas sobre o comportamento da pessoa, tanto diante de si mesma como diante das outras. A utilização de matérias-primas naturais orgânicas em formulações farmacêuticas e cosméticas segue uma tendência de mercado, haja vista o grande impulso que o mercado orgânico tem ocasionado na economia. Dentre estas matérias-primas, destaca-se a utilização do cacau orgânico que, pelo fato de ser rico em compostos polifenólicos e taninos condensados, acredita-se apresentar potencial ação antioxidante. Associados a sistemas nanoestruturados inovadores, como os sistemas líquido-cristalinos constituídos por silicones formadores de filme, a ação antioxidante do cacau orgânico pode, dessa forma, ser potencializada. Este trabalho tem como objetivos desenvolver e caracterizar sistemas líquido-cristalinos acrescidos de extrato de semente de cacau orgânico, avaliar a estabilidade físico-química e a atividade antioxidante das formulações obtidas. Realizou-se a caracterização do pó de cacau por microscopia óptica. A partir do pó do cacau, obteve-se extratos hidroalcóolicos do pó, a partir do qual foram quantificados compostos fenólicos utilizando o reagente Folin-Ciocalteu e determinada a atividade antioxidante com base na ação seqüestrante do radical 1,1-difenil-2-picrilhidrazila (DPPH). Phenolic compounds represent one of the main groups of secondary metabolites. Due to their chemical diversity, they have a variety of functions in plants, such as protection against ultraviolet radiation, herbivores and pathogens, and attraction of pollinators or dispersers of fruits and seeds. For human, the phenolic compounds are used like food colorings and flavors and, due to their pharmacological properties, mainly to antioxidant activity, they are associated with several health benefits, such as delay senility, prevention and therapy of cardiovascular diseases and of some cancers. The grapes are considered one of the main source of phenolic compounds and the fruit and its products are consumed in Brazil and worldwide. Considering the phenolic compounds diversity and their different distribution in the grape parts, this work had like aims the extraction, identification and quantification of the main classes of phenolic compounds of 10 grape varieties. The content of total phenols, flavanols and anthocyanins were determined, respectively, according to the Folin-Ciocalteu, DMACA and comparison of pH spectrophotometric methods. The content of total phenols varied from 142.75 + 1.86 to 483.39 + 5.90 mg.L-1 in the peel and from 86.50 + 0.54 to 146.32 + 9.97 mg.L-1 in the pulp. The amount of total flavanols varied from 3.68 + 0.03 to 6.92 + 0.26 mg.L-1 in the peel and from 0.90 + 0.00 to 1.36 + 0.00 mg.L-1 in the pulp. The content of total anthocyanins varied from 7.00 + 0.99 to 406.56 + 39.50 mg.L-1 in the peel and from 2.88 + 0.28 to 46.36 + 1.89 mg.L-1 in the pulp. The phenolic compounds concentration was higher in the peel than in the pulp. The total phenols and anthocyanins varied a lot while the total flavanols were more constant. The flavanols represent the smaller portion of phenolic compounds

The main objectives of this work were to evaluate the antioxidant activity and to determine of an ethanolic ginger extract total phenolic compounds concentration, as well as verifying its behavior by means of the oxidative stability, when added to refined soybean oil in concentrations of 0, 500, 1000, 1500, 2000 and 2500 mg/kg. The maximum antioxidant activity and EC50, concentration of the extract to achieve 50% of the antioxidant activity, value determined by free radical DPPH method were 79.1% and 42.6 g/mL, respectively. The total phenolic compounds concentration, determined to Folin-Ciocalteu method, was 251 mg/g. The induction period of the samples increased according to the augment of the extract concentration in the oil when evaluated using oxidative stability through the Rancimat equipament. It was possible to conclude that the ginger extracts possess effective action against the lipid oxidation and can be applied in foods as natural antioxidant.

Dentre as várias espécies de café, Coffea arabica L. destaca-se pela sua ampla importância comercial relacionada ao consumo da bebida e à produção de óleo. O grão de café possui grande quantidade de compostos fenólicos bioativos - destacando-se os com atividade antioxidante e fotoprotetora - e após a extração do óleo há sobra de grande quantidade de subprodutos, denominado torta. O objetivo desse trabalho é produzir um extrato a partir da torta de café selecionando o melhor solvente extrator, avaliando o teor de compostos fenólicos e o potencial antioxidante, para o emprego como ativo cosmético em formulações desenvolvidas, com análise in vitro para quantificação do Fator de Proteção Solar (FPS) dessas. Os extratos foram previamente produzidos por sonicação utilizando solventes de diferentes polaridades, e o extrato escolhido foi produzido em maior escala por maceração e realizado também extração exaustiva com o solvente escolhido (extrato etanólico 70%, v/v). Na determinação do teor de fenólicos totais foi utilizado o método com o reagente Folin-Ciocalteu. Foi avaliado também o rendimento (m/m) dos extratos e atividade antioxidante pelo método do radical DPPH. O extrato apresentou boa atividade antioxidante e alto teor de compostos fenólicos em comparação com outras plantas consideradas referências nessas propriedades. Para o preparo das formulações foram utilizados matérias primas aprovadas pela ANVISA, e as quantidades dos filtros solares químicos empregados para obtenção de Fator de Proteção Solar (FPS) teórico próximo de 30, utilizou-se como ferramenta o BASF - Sunscreen Simulator. As formulas desenvolvidas e preparadas foram analisadas com o equipamento Optometrics SPF 290S, capaz de fazer a determinação do FPS por análise in vitro. Phenolic compounds and antioxidant capacity are defense mechanisms of plants against the oxidative stress damage. Phenolic compounds are synthesized through the phenylpropanoid pathway, where the enzyme phenylalanine-ammonia-lyase plays a key role and it is influenced by light and photoreceptors such as phytochromes. The present research aims to evaluate the phenolic compounds content and antioxidant capacity of the wild Micro-Tom (MT) cultivar tomato fruits and its photomorphogenic mutant tomato plants high pigment 1 (hp1), super responsive to events mediated by light, and aurea (au), quantitative phytochrome deficient. Twenty mature fruits of each genotype (MT, hp1, au) were used in triplicate for analyses. To quantify the total phenolic compounds the Folin-Ciocalteu method was used and the antioxidant capacity was analyzed by Ferric Reducing Antioxidant Power (FRAP) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) methods. The hp1 mutant presented the highest total phenolic compounds content and higher antioxidant capacity than wild cultivar (MT) and au mutant, which did not differ significantly from MT cultivar.
